چکیده

A mutually noncomplementing group of phage P22 mutants (L mutants) have been isolated which cannot accomplish a late step necessary for prophage integration. L mutants produce turbid plaques and lead to cell survival on high multiplicity infection, but the infecting phage genomes are incapable of integrating as prophage and are progressively diluted among the segregating cells. L mutants can become prophage by complementation with L+ phage. The L+ gene apparently produces a diffusible product which acts lrans to accomplish integration. Experiments with temperature sensitive L mutants show that the gene begins acting late, after the infected cells have recovered and begun to divide, and does not accomplish its effect in all the cells until at least the third generation. L lysogens formed by complementation are stable. The L+ gene is thus necessary for integration but not for maintenance of the lysogenic state.

Specialized Transducing Phages Derived from Salmonella Phage

Salmonella phage P22 has been used in the construction of three sorts of specialized transducing phage: P22 proAB, P22 proABlac and P22 argF. The bacterial genes carried are derived from E. coli K12. Since E. coli and Salmonella chromosomes recombine very poorly, E. coli genes cannot be transduced into Salmonella recipients by P22‘s generalized transduction mechanism. Therefore, stable inherita...

Transduction by phage P22 in a recombination-deficient mutant of Salmonella typhimurium.

Recombination-deficient (Ret-) mutants of Salmonella typhitn&um were used to study special transduction by phage P22 in the absence of the usual background of general transduction. Retstrains could not be transduced for any marker with a lysate arising by infection. When the same strains were infected with a lysate obtained by induction, low but significant levels of transduction were observed ...

Defective Phage Formation by Lysogens of Integration

Integration deficient (L) mutants of phage P22 can be complemented with L+ phage to yield L mutant lysogens. Once established the L prophage is stable in the absence of L+ function. Ultraviolet or thermal induction of L lysogens leads to product, ion of defective phage particles and low yields of infectious phage. The defective particles contain a normal amount of DNA but the DNA is mainly bacte...

Regional specificity of illegitimate recombination by the translocatable ampicillin-resistance element Tn1 in the genome of phage P22.

Insertions of the translocatable ampicillin-resistance element Tn1 were selected in the genome of the temperate Salmonella phage P22 by growing the phage on hosts carrying the resistance plasmid RP4. Insertions of Tn1 into phage P22 are rare (10(-10) per phage) and nonrandomly distributed in the P22 genome. They are found mainly in the vicinity of the P22 ant gene. Insertions within the ant gen...

Genetics of bacteriophage P22. I. Isolation of prophage deletions which affect immunity to superinfection.

Two kinds of prophage deletions of the Salm.onella phage P22 were isolated. One type of deletion was isolated by simultaneously selecting for loss of a host gene (pro) adjacent to the integrated prophage and for survival at the nonpermissive temperature of a temperature-inducible (tse2) lysogen. The second type of deletion was isolated by transduction to tetracycline resistance using high-frequ...
